For anyone facing a similar issue, I found the hint for my solution in

It appears to happen with MacOS Sequoia, if the DNS does not support 
EDNS0 packets.
In this case the option "Limit IP address tracking" needs to be disabled 
for each network interface, which in my case was "on" by default.

Can the EDNS0 option be supported via dnsmasq?
